Traditional antibody discovery methods, such as hybridoma technology and B cell screening using platforms like flow cytometry and ELISA, are constrained by limited repertoire screening capacity and focus primarily on binding assays. This restricts the functional understanding of candidates, including their blocking and neutralizing properties, cross-reactivity, and immunogenic effects. Here we present the Lightcast platform that leverages droplet microfluidics to conduct highly controlled, sequential, and multiplexed single-cell assays in parallel workflows, enabling a deeper understanding of antibody function.
